Title: Models of RNA recognition and Processing by Pontocerebellar Hypoplasia Linked Nucleases
Short Abstract:聽RNAs throughout our cells require proper processing to reach their functional, mature forms. The lab is interested in understanding the ways in which different RNA processing enzymes are able to recognize their RNA targets, through biochemical specificity and cellular regulation. I will present recent work on two RNA processing complexes, the tRNA Splicing Endonuclease Complex (TSEN) and the Target of EGR1 (TOE1), which are linked through unknown mechanisms as mutations in both RNA processing complexes cause Pontocerebellar Hypoplasia (PCH). We will share work in which we have helped uncover how the TSEN complex recognizes tRNAs, developed novel insights into the regulation splicing by the TSEN accessory protein, CLP1, and most recently demonstrated the role of PCH linked mutations in interrupting TOE1 function.
